ABSTRACT

Chronic obstructive pulmonary disease (COPD) is a common disease in clinical practice. It is associated with obvious exposure to toxic particles or gases and has become the leading cause of death and disability worldwide. The pathogenesis of COPD is complex, and the oxidative stress involved in COPD plays a crucial role in the pathological process of the disease. Patients with COPD usually have high levels of oxidative stress in the lungs, which will affect the whole body for a long time, causing a variety of complications and accelerating the development of the disease. On the one hand, oxidative stress can directly damage the airway and lung tissue. On the other hand, it also drives other pathological mechanisms to jointly promote the development of disease, such as participating in inflammatory reactions and protease/anti-protease imbalance, promoting mucus secretion, accelerating cellular senescence, causing autoimmunity, and involving in genetic regulatory pathways. At present, western medicine treatment is mostly based on conventional drug treatment, and antioxidant-targeted oxidative stress is adopted, but there are still some challenges in efficacy and safety. Traditional Chinese medicine has a long history of preventing and treating COPD. In particular, Chinese herbal medicine formulas have great potential to interfere with the oxidative stress of COPD. Whether it is the modified classical traditional Chinese medicine or the new formulation developed by modern doctors, the research results reflect the multi-target and multi-channel advantages of traditional Chinese medicine treatment, and their efficacy and safety are gradually verified. This paper reviewed the literature in recent years, starting with the basic and clinical research on the intervention of traditional Chinese herbal medicine formulas on oxidative stress of COPD, so as to provide further ideas for related research on the prevention and treatment of oxidative stress of COPD by traditional Chinese medicine.

ABSTRACT

Chronic obstructive pulmonary disease (COPD) is characterized by long treatment course and poor prognosis. The pathogenesis has not been fully elucidated but is mostly related to the non-specific inflammation of the airway and surrounding tissues. T helper 1 (Th1) and T helper 2 (Th2) are generated by CD4+ T cell differentiation, and are in a dynamic equilibrium when the body is in normal state. The balance between pro-inflammatory cytokines and anti-inflammatory cytokines regulated by Th1/Th2 is vital for maintaining the immune homeostasis in respiratory tract. Chronic inflammatory state changes the level of inflammatory cells in the body, and there is immune disorder in T lymphocytes in the onset stage of COPD. Th1 cells are predominantly expressed in the stable stage of COPD, while Th2 cells are predominantly expressed in the acute exacerbations of COPD (AECOPD). Th1/Th2 immune imbalance aggravates the inflammatory reaction, and thus restoring the immune balance between them and inhibiting the inflammatory reaction are critical for the treatment of COPD. At present, there has been no satisfactory treatment plan for COPD. Chinese medicine has a long history of preventing and treating COPD, with remarkable curative effect and few adverse reactions. A large number of animal experiments and clinical studies on Chinese medicine intervention of Th1/Th2 immune balance in COPD have indicated that Th1/Th2 immune balance is an important potential target for treating COPD by Chinese medicine, which can correct chronic inflammatory state by regulating the immune disorder of the body. It has also been found that Th1/Th2 balance plays an important immunoregulatory role in inflammatory response, but little is known about its specific mechanism in the pathogenesis of COPD. On this basis, this paper summarized and analyzed the biological characteristics of Th1/Th2 and their mechanism in the pathogenesis of COPD, as well as the intervention effect of single Chinese medicine or its effective components and Chinese medicine compound on Th1/Th2 immune balance in COPD. It further explored the pathogenesis of COPD and the potential therapeutic targets of Chinese medicine in interfering with Th1/Th2 immune balance in COPD, providing reference for further study on prevention and treatment of COPD with Chinese medicine.

This article analyzed, organizde, and summarized the specific meaning of yin and yang in Shang Han Lun, holding the idea that Zhongyang in Shang Han Lun contains fluid. YE Tian-shi's Tong Yang Bu Zai Wen Er Zai Li Xiao Bian is derived from Zhong jing method, and is more suitable to be used in damp-heat syndromes, reflecting that YE Tian-shi is not confined to traditional methods, but follows traditional prescriptions and flexible thoughts, which provides references for learning and flexibly applying prescription ideas.

ABSTRACT

Objective To investigate the mechanism of Guiqiyiyuan Ointment (GO) for preventing and treating 12C6+ beam radiation induced lung and kidney bystander effect to provide a new strategy for prevention and treatment of clinical radiation injury. Methods Sixty healthy male Wistar rats were randomly and equally divided into 3 groups: NC group, SR group (simple radiation 2ml/kg), GO group(GO 2ml/kg intragastric administration for 7 days). The right side of the lung was modeled by 12C6+beam radiation. After modeling, the rats were killed at 48h. The left lung, left and right kidney tissues were taken from the rats. The DNA methylation rate was detected by ELISA assay, pathological changes were observed by HE staining, and the expressions of Dnmt1, Dnmt3a and Dnmt3b were detected by immunohistochemistry. Results Compared with the NC group, the level of DNA methylation was decreased significantly (P<0.01), the left lung showed inflammation, no abnormal finding was seen in the left and right kidneys, and the expressions of Dnmt1, Dnmt3a and Dnmt3b were significantly increased in the SR group (P<0.01). Compared with the SR group, the level of DNA methylation was increased significantly (P<0.01), the left lung inflammation became better, and the expressions of Dnmt1, Dnmt3a and Dnmt3b were significantly decreased in the GO group (P<0.01). Dnmt1, Dnmt3a and Dnmt3b proteins were expressed in the cytoplasms of bronchial and renal tubular epithelial cells in all the groups. The NC group presented as light brown-brown staining, showing a weak positive expression, the SR group as brown-brown staining, showing astrong positive expression, and the GO group as light brown-brown staining, showing a moderate positive expression. Conclusion The GO can reduce the bystander effect caused by 12C6+ beam radiation, and its mechanism is related to improving the level of DNA methylation.

ABSTRACT

Objective To discuss the protective effects ofGuiqi YiyuanOintment on damages caused by heavy ion radiation induced bystander effects; To explore the possible mechanism.Methods Totally 42 Wistar male rats were randomly divided into blank control group, pure radiation group andGuiqi Yiyuan Ointment group. TheGuiqi Yiyuan Ointment group was givenGuiqi Yiyuan Ointment by gavage for two weeks in advance. Later the right lungs of the rats in the pure radiation group andGuiqi Yiyuan Ointment group were radiated once by 2 Gy12C6+, while the blank control group received no radiation. 6, 12, 24 h after radiation all groups of rats were executed. Peripheral hemogram and the levels of IL-6, TGF-β1 and IL-1β in serum of the rats were examined. The changes of lung tissue pathology morphology in the rats were observed by HE staining.Results Compared with the blank control group, the contents of IL-6, TGF-β1 and IL-1β in serum of the pure radiation group increased obviously at 12 h after radiation (P<0.01), and the amount of leukocyte, erythrocyte, blood platelet and hemoglobin distinctly declined at 12 h after radiation (P<0.01); HE staining showed that the alveolar wall was thickened at 24 h after radiation, and there were exudate and inflammatory cell infiltration in the alveolar cavity. Compare with the pure radiation group at 12 h after radiation, the levels of IL-6, TGF-β1 and IL-1β inGuiqi Yiyuan Ointment group decreased significantly at 12 h after radiation (P<0.01). Indexes of blood routine significantly increased (P<0.01), and the pathological changes of lung tissue in rats improved (P<0.01). ConclusionGuiqi Yiyuan Ointment can protect damages caused by heavy ion radiation induced bystander effects.

ABSTRACT

OBJECTIVE:To establish the technical system that is suitable for protein extracting in Angelicae sinensis seed and sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE),and provide technical support for detecting the protein quality and variety purity. METHODS:Using protein content and number of electrophoretic bands as indexes,8 methods,includ-ing voncentrated gel method,salt-soluble protein method,electrode buffer method,dimercaptosyl alcohol (DTT) method,urea method,mercaptoethanol method,trimethylolaminomethane (Tris) method,and acetone precipitation method,were conducted to extract the protein in A. sinensis seed and screen the optimal extraction method. Then based on optimal extraction method,effects of different materials-lipid ratios,sample dilution times(sample volume)and separate gel concentration on SDS-PAGE were investi-gated. RESULTS:Mercaptoethanol method extracted the highest protein contents(29.931 mg/g),with many electrophoretic bands and clear background. When mercaptoethanol method was used as optimal extraction method,electrophoresis effects were the best in the conditions of materials-lipid ratio of 1:10,sample volume of 5 times,separate gel concentration of 15%,which obtained 18 bands totally. CONCLUSIONS:Established protein extraction method and SDS-PAGE technical system are suitable for detecting the purity of A. sinensis seed.

ABSTRACT

OBJECTIVE:To select the optimum conditions for germination test of Sophora alopecuroides seed,and to provide reference for formulating seed test rule and standardization of S. alopecuroides. METHODS:S. alopecuroides seeds were soaked in 98% H2SO4 for 30 min and 35 ℃ warm water for 29 h,and then treated under different temperature conditions (15 ℃,20 ℃, 25℃and 30℃),different germination beds(on paper,between paper,on sand,in sand)and different light conditions(2 000 lx lighting 16 h,dark). Optimal germination condition was screened by using germination rate,germinative potential and germinative index as indicators. S. alopecuroides seeds were cultured under this condition for 7 days,and then germination rate was determined. RESULTS:Different temperatures had no significant effect on germination rate,but influenced germinative potential and germina-tive index;those indicators reached maximal value at 20 ℃. Different germination beds affected each indicator,and under condi-tion of on sand,those indicators were the highest. Light treatment had no significant effect on germination indicators. Under suit-able condition,the seed sprouted since first day of germination bed treatment;germination rate was more than 90% on second day,and reached maximal value on fifth day and didn’t increase any longer. CONCLUSIONS:The suitable condition of seed ger-mination was soaking in 98% H2SO4 30 min+35 ℃ warm water for 29 h,on sand under light at 20 ℃. Initial count on second day of germination bed treatment and final count on forth day were analyzed statistically as well as germination rate. This method can be used as standard quality test of the seed of S. alopecuroides.

ABSTRACT

OBJECTIVE:To select the optimum conditions for germination test of Astragalus membranaceus seed and provide reference for the formulation of testing rules of A. membranaceus seed. METHODS:First,the dormancy of A. membranaceus seeds were broken by soaking seed with 98% H2SO4 for 30 min and 35 ℃ warm water for 9 h,then treated with different temperatures of germination,different germinating beds and light conditions. Different treatment methods were evaluated by germination rate, germination potential and germination index. RESULTS:With the increase of temperature,germination rate,germination potential and germination index decreased. At 30 ℃,germination rate,germination index,and germination potential were significantly low-er than those of seed with other treatments,in which the germination rate was the highest at 15℃;but there wasn’t significant dif-ference in germination rate of 15 ℃ and 20 ℃. There were no significant differences in germination rate of different germination beds,but sand bed could restrain bacterial growth well. Under both light and dark conditions,seed could normally germinate. The appropriate condition of A. membranacus seed germination was at 15 ℃,sand bed and dark. The germination rate,germination po-tential and germination index were(98.5±0.65)%,(85.5±0.87)%and 175.8±2.31,respectively. Meanwhile,the germination pe-riod was only 4 days. CONCLUSIONS:Suggested quality control method of A. membranaceus seed is that at 15 ℃,sand bed and dark. The second day of germination as initial counting time and forth day as the last counting time are used to calculate germina-tion index. This method is easy and controllable. It also has short germination period and high germination rate.

ABSTRACT

Objective To observe the effects of Radix Hedysari on collagen area, hyaluronic acid (HA) and laminin (LN) of lung tissues in rat with pulmonary interstitial fibrosis; To investigate the mechanism and screen the best active ingredients in Radix Hedysari.Methods Totally 144 SPF Wistar rats were randomly divided into control group, model group, metacortandracin group, and Radix Hedysari polysaccharide, flavone and saponin groups were set as high-, medium-, and low-dose groups. Pulmonary interstitial fibrosis models were set up by dropping bleomycin into air tube of rats. All groups were taken corresponding medicine with appropriate dose daily on day 2 after models were established for 28 days. Collagen fibrils in lung tissue were observed by Masson dying and contents of HA and LN in lung tissue of rats in each group were detected by radioimmunoassay.Results Compared with the model group, pulmonary alveoli in Radix Hedysari flavone high-, medium-, and low-dose groups and Radix Hedysari polysaccharide low-dose group were relieved obviously; collagenous fiber area in Radix Hedysari flavone low-dose group, Radix Hedysari polysaccharide medium-dose group, and Radix Hedysari saponin low-dose group decreased obviously. Compared with the model group, the content of HA in Radix Hedysari polysaccharide high-dose group, Radix Hedysari flavone high-, medium-, and low-dose groups, and Radix Hedysari saponin high-dose group decreased obviously; the content of LN in Radix Hedysari polysaccharide high-dose group, Radix Hedysari flavone medium- and low-dose groups, and Radix Hedysari saponin medium- and low-dose groups decreased obviously. Conclusion Polysaccharide, flavone and saponin of Radix Hedysari have the abilities to alleviate inflammation of pulmonary alveoli, inhibit proliferation and deposition of collagen fibrils, and reduce the contents of HA and LA in lung tissue. Among these active ingredients, flavone has the best effect.

ABSTRACT

Objective To discuss radix hedysari flavonoids mechanism of preventing pulmonary fibrosis.Methods Totally 216 SPF Wistar rats were randomized into normal group, model group, prednisone group, radix hedysari flavonoids high, medium, and low dose groups, and then pulmonary fibrosis model was established by intratracheal dripping of bleomycin. From the second day after modeling, every treatment group received gavage with the corresponding dose of medicine at the planned time for 7, 14, and 28 continuous days. Expressions of MMP-2 and TIMP-1 protein were detected by immunohistochemical method.Results When radix hedysari flavonoids high dose group was at the 7th, 14th, 28th day, and medium dose group was at the 14th day, MMP-2 protein expression was lower than model group, similar to prednisone group. When radix hedysari flavonoids high dose group was at the 14th, 28th day and medium dose group was at 14th day, TIMP-1 protein expression was lower than model group.Conclusion Radix hedysari flavonoids can adjust the expressions of MMP-2 and TIMP-1 protein at different phases, and tend to make the balance of MMPs/TIMPs, which may be an effective mechanism for the inhibition of fibrosis process.

ABSTRACT

Objective To investigate effect of peripheral blood miR-21 expression on the clinicopathological features of gastric cancer before and after gastric surgery.Methods The peripheral blood was collected on the operative day and postoperative 7 d in 42 patients with primary gastric cancer surgery.The real-tine quantitative PCR was adopted to detect the relative expression of miR-21 in plasma with U6 as an internal reference gene.Results The plasma miR-21 expression level after gastric cancer operation was significantly down-regulated compared with before operation(P <0.01),the overall relative expression level was dropped by an average of 18.2 times;and it was found that the postoperative miR-21 expression in the patients with the tumor family history was up-regulated,the postoperative plasma miR-21 expression in the patients without the tumor family history was decreased by 22 times compared with before operation(P <0.05);the experimental results also fund that as the patient′s differentiation degree was changed from high to low,the lymph node metastasis rate was gradually increased,while postoperative average miR-21 expression was changed from low to high,showing the relative expression times of miR-21 were distributed in the four differentiated zones of high,middle,middle-low and low,which showed the correlation with the distribution of the lymph node metastasis rate in these four differentiation zones.Conclusion The peripheral blood MiR-21 detection has the clinical significance to early auxiliary diagnosis and the judgement of postoperative malignant degree.

ABSTRACT

Objective To discuss the effects of Radix Hedysari flavonoids on microvessel density (MVD) of pulmonary interstitial fibrosis model rats, and provide evidence for its development. Methods SPF Wistar rats were divided into blank group, model group, prednisone group, and Radix Hedysari flavonoids high-, medium- and low-dosage group. Pulmonary interstitial fibrosis model was established by intratracheal instillation of bleomycin. From the second day after model established, each drug treatment group was administered with corresponding drugs intragastrically at different points in time for 14 and 28 days. MVD was detected by immunohistochemistry. Results Neomicrovessel was increased significantly in pulmonary tissue of model rats of 14 days treatment, but slightly decreased in 28 days. MVD in 14, 28 days model group was significantly more than blank group (P<0.01). MVD in 14, 28 days Radix Hedysari flavonoids high dosage group was significantly less than model group (P<0.01). MVD in Radix Hedysari flavonoids medium-dosage group was between high-and low-dosage group, and MVD in medium-dosage group was similar with that in model group. Conclusion Radix Hedysari flavonoids could inhibit the formation of neomicrovessel in a dose dependent manner.

ABSTRACT

Objective To observe the effects of Hedysari flavonoids on TGF-β1 and ultrastructure in lung tissue of rats with pulmonary fibrosis, and explore its mechanism. Methods Wistar rats were randomly divided into blank group, model group, prednisone group and Hedysari flavonoids high-, medium- and low-dose groups. Pulmonary fibrosis model was established by injection of bleomycin via trachea. After 24 h, rats were given drugs by gavage in all treatment groups once daily. The expression of TGF-β1 in lung tissue was detected by immunohistochemistry on d7, d14 and d28, and ultrastructure wasobserved by transmission electron microscope on d28. Results The positive expression of TGF-β1 was found in a few bronchiolar epithelial cell cytoplasm of blank group. Compared with the blank group, the positive expression of TGF-β1 on bronchial epithelial cell significantly increased (P<0.05) on d7, d14, and d28 in model group. Compared with the model group, the positive expression of TGF-β1 were significantly decreased (P<0.05, P<0.01) in Hedysari flavonoids high- and medium-dose groups and prednisone group on d7, d14 and d28. With electron microscopy, collagen fibers within the mediastinum in rat alveolar in model group were significantly increased, cell cytoplasm mitochondria was swollen in type Ⅱ epithelial cell.Mitochondrial crista was broken or disappeared, lamellar bodies significantly reduced. Collagen fibers reduced significantly in all treatment groups, which can improve and change ultrastructure on type Ⅱ epithelial cell. Conclusion Hedysari flavonoids can significantly reduce pathological damage and extracellular matrix deposition of rats with pulmonary fibrosis, which may have connection with inhibiting of TGF-β1 expression.

ABSTRACT

Objective To observe the effects of Qizhi Zhoufei Granules on IL-8, TNF-αand ET-1 in serum and bronchoalveolar lavage fluid (BALF) of chronic obstructive pulmonary disease (COPD) model rats, and to study its mechanism of preventing and treating COPD. Methods Wistar rats were randomly divided into blank group, model group, positive control group, Qizhi low-dose group, Qizhi medium-dose group and Qizhi high-dose group. The COPD model was established by smoking and infusing lipopolysaccharide (LPS). After 15 days, the rats were given the drugs by orally taking once a day. On the 43rd day, the change of IL-8, TNF-α and ET-1 content in serum and BALF were measured by radio-immunity assay. Results Compared with the blank group, the level of IL-8, TNF-α and ET-1 in serum and BALF were considerably raised in model group (P<0.05). Whereas compared with the model group, the level of TNF-αin serum of Qizhi high-dose group was remarkably lowered (P<0.05), the level of IL-8 in serum of Qizhi low-dose group and Qizhi high-dose group was remarkably lowered (P<0.05), the level of TNF-αin BALF of Qizhi medium-dose group was remarkably lowered (P<0.05), the level of IL-8 in BALF of Qizhi low-, medium- and high-dose group was remarkably lowered (P<0.05), and the level of ET-1 in BALF of Qizhi high-dose group and Qizhi medium-dose group was remarkably lowered (P<0.05). Conclusion Qizhi Zhoufei Granules have protective effect on lung tissue of COPD rats, and can adjust the content of IL-8, TNF-α and ET-1, which may be one of its mechanisms of intervening occurrence and development of COPD.

Objective To observe the effect of Pingchuanlingchongji (PCL) on the auricular microcirculation of mice and to investigate the effect mechanism of the treatment for asthma. Methods 50 mice were randomly divided into five groups, group 1 and group 2 were given 0.9% NS (25 mL/kg) as control group; group 3 was given aspirin (0.25 g/kg), 4 group was given low-dose (1.1 g/kg) PCL, group 5 was given high-dose (2.2 g/kg) PCL. Last for seven days. We injected Posterior Pituitary Injection (0.5 U/mL) on the rear vein of 2-5 group mices after the last time of given druy. The diameter of micro-artery in mice auricular, the diameter of micro-vein and blood flow speed were observed. ABSTRACT

<p><b>BACKGROUND</b>To study the inhibitive effect of LAK cells induced by dendritic cells (DCs) on implanted lung adenocarcinoma in nude mice.</p><p><b>METHODS</b>The lung adenocarcinoma model was constructed in nude mice using the resected samples of lung cancer patient. The lung cancer cell lysate was obtained by free-zing and thrawing cycles. Peripheral blood mononuclear cells (PBMNC) were obtained from venous blood of the same patient, in which the adherent PBMNC fraction was cultured with DCGF, and the non-adherent PBMNC fraction was cultured with rhIL-2. DCs were pulsed with lung cancer cell lysates. And then mature DCs were incubated with LAK cells and the mixed cells were named DC-LAK cells. DC-LAK cells were injected into lung cancer-bearing nude mice to observe the inhibitive effect.</p><p><b>RESULTS</b>The lung adenocarcinoma mo-del was successfully constructed. The average tumor weights of DC-LAK, LAK, DC and saline control groups were 0.47, 1.05, 1.30 and 1.58 g respectively, and the inhibitive rates of DC-LAK, LAK and DC were 70.3%, 33.5% and 17.9% respectively. The antitumor activity of DC-LAK cells was significantly stronger than that of LAK cells (P < 0.05).</p><p><b>CONCLUSIONS</b>The results of in vivo experiment show that the antitumor activity of DC-LAK cells is stronger than that of LAK cells, so DC-LAK cells treatment may be a more efficient approach of lung cancer biological therapy. This experiment may provide a foundation for clinical application of DC vaccine.</p>

<p><b>BACKGROUND</b>To investigate the distribution of TCR Vβ subfamily T clonal cells in peripheral blood lymphocytes (PBL), tumor infiltrating lymphocytes (TIL) and lymphocytes in non-cancerous lung tissues of patients with non-small-cell lung cancer (NSCLC) and to see the inclination of the T cell antigen receptor (TCR) Vβ subfamilies' expression.</p><p><b>METHODS</b>Complimentarily determining region 3 (CDR3) of TCR 24 variable region genes was analyzed in PBL, TIL and lymphocytes in non cancerous lung tissues from 24 NSCLC cases with reverse transcriptase-polymerase chain reaction (RT-PCR) and gene scan techniques to identify the distribution and clonality of TCR Vβ subfamily T cells.</p><p><b>RESULTS</b>Only a portion of Vβ T cells were found in patients with NSCLC, whereas 24 TCR Vβ subfamily T cells were detected in 10 healthy controls. Vβ5 subfamily was expressed mostly in TIL and the frequency of Vβ5 in TIL (6/18, 33.3%) was much higher than that of PBL (1/24, 4.2%) and T cells infiltrating non-cancerous lung tissues (0/12) (P < 0.05). Oligoc lonal T cells were found in 2 cases with Vβ5 subfamily and polyclonal T cells in 4 cases.</p><p><b>CONCLUSIONS</b>There are dominant and clonal TCR Vβ subfamilies expressed in TIL of NSCLC patients, which may be the tumor associated antigens (TAA) specific.</p>

ABSTRACT

<p><b>BACKGROUND</b>To culture and proliferate dendritic cells from the peripheral blood of patients with lung cancer, to observe the morphologic and phenotypic characteristics of these cells and to lay a foundation for applying dendritic cells in the biotherapy of lung cancer.</p><p><b>METHODS</b>The mononuclear cells isolated from peripheral blood of patients with lung cancer were cultured with the supplement of DCGF to generate dendritic cells. Tumor associated antigens were added into dendritic cell culture on the eighth day. The morphologic characteristics of those cells were observed with inverted or electron microscopes and the phenotypic figures were analyzed with FCM.</p><p><b>RESULTS</b>A large number of mature dendritic cells were obtained from the peripheral blood of patients with lung cancer and showed specific morphologic and phenotypic characteristics. The specific antigen expressed as follows: CD80: 81.8%, HLA DR: 98.3%, CD86: 69.8%, CD1a: 19.7%, CD14: 83.1%.</p><p><b>CONCLUSIONS</b>The mature dendritic cells can be obtained by culturing the peripheral blood of lung cancer patients, which lays the further foundation for clinical biotherapy of tumor.</p>

ABSTRACT

<p><b>BACKGROUND</b>To explore the correlation of microvessel density (MVD) and microvessel structure (MVS) features with the patients' prognosis in non-small cell lung cancer (NSCLC).</p><p><b>METHODS</b>Anti-Von Willebrand factor antibody was used to stain microvessel endothelia by means of LsAB immunohistochemical technique, then the microvessel count and structure features were observed microscopically in 49 primary NSCLC tissues. MVS pattern A had scattered microvessels with relatively integral or thick wall and with relatively regular morphology and MVS pattern B had plexiform or network like microvessels with unintegral or thin wall and with irregular morphology.</p><p><b>RESULTS</b>MVD in primary NSCLC tissues was closely correlated with pTNM stage or lymph node involvement, P=0.043 and P=0.038, respectively. MVS in primary NSCLC tissues was closely correlated with the size of primary carcinoma, P=0.002. The survival of patients (23.2± 18.4 months) with MVD > 52/200× was significantly shorter than that of patients (35.9±20.9 months) with MVD < 52/200× in primary NSCLC tisssues, P=0.01. The survival of patients with MVS pattern A (39.4±17.2 months) was significantly longer than that of patients with MVS pattern B (23.5±20.3 months) in primary NSCLC tisssues, P=0.008. The survival of patients with MVD < 52/200× and MVS pattern A (42.9±19.3 months) was significantly longer than that of patients with MVD > 52/200× and MVS pattern B (15.7±16.8 months) in primary NSCLC tissues, P=0.002.</p><p><b>CONCLUSIONS</b>MVD and MVS are closely associated with prognosis of NSCLC patients and might be served as parameters estimating patients' prognosis and planning assistant therapy after operation.</p>

ABSTRACT

Purpose:To investigate the relationship between inactivity of mismatch repair system in colorectal cancer (CRC) in Chinese patients and their age at onset of CRC.Methods:Genomic DNA extracted from colorectal cancer tissues and their normal colon tissues were subjected to analysis for microstallite instability (MSI) in six of DNA markers in 100 colorectal cancer patients.Results:Microsatellite instability positive (MSI + ) was detected in 46 out of 100 (46%) of CRC tissues, MSI + H in 18/100 (18%) and MSI + L in 28/100 (28%).
